Abstract
Purpose
: To evaluate a suitable material for in vivo cell therapy and tissue engineering purposes, we have tested biodegradable materials, such as polymer and helistat. Materials and Methods : We injected chondrocytes and fibroblasts with or without TGF-β secreting cells into two types of polymer and helistat, and then injected into mouse peritoneum. The chondrocytes were cultured from patient's knee joint cartilage with degenerative arthritis. TGF-β secreting cells were prepared by calcium phosphate transfection and the TGF-β secretion was measured by ELISA assay. Balb/c mouse was used for in vivo culture. Results : At 6 weeks after injection into helistat, the various cell types with additional TGF-β secreting cells made tissues which were derived from the injected cells. Control cells without TGF-β secreting cells made scanty amount of tissues. Conclusion : Cell therapy or tissue engineering approaches with helistat can be a very useful method for the purpose of tissue regeneration, especially, when they are carried out with additional gene therapeutic approach.
