Steroid Induced Apoptosis in Rabbit Achilles tendon and NIH 3T3 , Foreskin Fibroblasts

Abstract
To evaluate the safety of repeated steroid injection in vivo in relation to apoptosis, we did staining for apoptosis and flow cytometric analysis in rabbit achilles tendon and NIH 3T3, foreskin fibroblasts after exposure to steroid. Three different concentrations of steroid were exposed to NIH 3T3 cells and foreskin fibroblasts. Rabbit achilles tendons were also injected with steroid serially. Apoptosis was not observed in most of the concentrations of steroid in NIH 3T3 cells and foreskin fibroblasts in vitro. Apoptosis was not found in NIH 3T3 cells and foreskin fibroblasts with flow cytometric analysis. But in repeated injections of steroid to rabbit achilles tendons, apoptosis was observed. We concluded that even though single dose of steroid is safe from inducing apoptosis, care must be taken for the repeated injections of steroid because it can induce apoptosis in vivo.
